چکیده

Endocytosis by mammalian cells leads to the rapid uptake of exogenous solutes within plasma-membrane-derived vesicles. In general, fusion of these vesicles with primary or secondary iysosomes occurs and most biological macromolecules are degraded to small molecular weight components (1). Extensive bidirectional flow of membrane takes place between the plasma membrane and the lysosomal apparatus during endocytosis (7). Recent evidence has suggested that solutes pass through at least one intracellular compartment before reaching the lysosome (26) and representative portions of endocytic markers (2, 4, 6, 8) are returned to the cell surface. We now report the existence of a distinct, intermediate vacuolar compartment in the parasitic amoeba, Entamoeba histolytica. This is the primary pinocytic route in this organism. It is equilibrated with the pH of the ambient milieu and allows for the flow of solutes into and out of the cytoplasm. Only after many hours of intraceilular residence are the solutes detected in a separate, acidified lysosomelike compartment.

Endocytosis in Entamoeba histolytica. Evidence for a unique non- acidified compartment

Our studies on endocytosis in Entamoeba histolytica trophozoites suggest that there are two vacuolar compartments in this organism. The first compartment consists of large vacuoles (greater than 2 microns diameter). As measured by the fluid phase markers, fluorescein isothiocyanate (FITC)-dextran and horseradish peroxidase (HRP), this compartment is a rapid equilibrium with the external milieu ...

Data on docking and dynamics simulation of Entamoeba histolytica EhADH (an ALIX protein) and lysobisphosphatidic acid

Entamoeba histolytica is the protozoan agent responsible for human amoebiasis. Trophozoites are highly phagocytic cells and the lysobisphosphatidic acid (LBPA) is involved in endocytosis. LBPA interacts with EhADH protein (an ALIX family member) also participating in phagocytosis, as it is referred in the research article Identification of the phospholipid lysobisphosphatidic acid in the protoz...

Identification of the phospholipid lysobisphosphatidic acid in the protozoan Entamoeba histolytica: An active molecule in endocytosis

Phospholipids are essential for vesicle fusion and fission and both are fundamental events for Entamoeba histolytica phagocytosis. Our aim was to identify the lysobisphosphatidic acid (LBPA) in trophozoites and investigate its cellular fate during endocytosis. LBPA was detected by TLC in a 0.5 Rf spot of total lipids, which co-migrated with the LBPA standard. The 6C4 antibody, against LBPA reco...

The cultivation of entamoeba histolytica in HSr+ s medium

In 1926 for the first time Dobell and laid low synthesized and used Hsre+ s ( horse serum, ringer, egg+ starch rice) as a culture medium for growth of E.histolytica. since then this medium along with tens of other media have been used for growth of E.histolytica and other intestinal microorganisms. This medium has been used worldwide in many diagnostic and research laboratories. In Iran, in ...

Involvement of raft-like plasma membrane domains of Entamoeba histolytica in pinocytosis and adhesion.

Lipid rafts are highly ordered, cholesterol-rich, and detergent-resistant microdomains found in the plasma membrane of many eukaryotic cells. These domains play important roles in endocytosis, secretion, and adhesion in a variety of cell types. The parasitic protozoan Entamoeba histolytica, the causative agent of amoebic dysentery, was determined to have raft-like plasma membrane domains by use...
